VoriconazoleNPharmacokineticsNin Immunocompromized Children Voriconazole is increasingly used in pediatric patients, but only a few studies have reported on the pharmacokinetics in this population. Important pharmacokinetic differences exist between adults and children receiving voriconazole, primarily a higher elimination capacity in pediatric patients, likely due to their greater ratio of liver mass to body mass than seen in adults. An open-label, parallel-group study of the pharmacokinetics and safety was done in immunocompromized children aged 2 to less than 12 years and adults. Twelve children and 12 adults with hematological malignancies who required treatment for the prevention or therapy of systemic fungal infections were included in the study. Nine children received a voriconazole dose of 7 mg/kg intravenously every 12 hours over the entire study period. Twelve adults and three children received two loading doses of 6 mg/kg every 12 hours, followed by a maintenance dose of 5 mg/kg for the children or 4 mg/kg for the adults, both given every 12 hours. The children had mean age of 7 years and the adults averaged 44 years of age. The plasma trough levels of voriconazole in children receiving 7 mg/kg were not significantly different from the values obtained in adults receiving a dose of 4 mg/kg intravenously. The children had significantly higher C max values, but other pharmacokinetic parameters were not significantly different from those of adults. Voriconazole exhibited nonlinear pharmacokinetics in the majority of children. In children 2 to < 12 years old, the European Medicines Agency approved dose of 7 mg/kg every 12 hours of voriconazole intravenously provides exposure (area under the curve) comparable to that observed in adults with a dose of 4 mg/kg every 12 hours. Diagnosis and treatment of superficial vein thrombosis.

Superficial vein thrombosis (SVT) is a common disease, characterized by an inflammatory-thrombotic process in a superficial vein. Typical clinical findings are pain and a warm, tender, reddish cord along the vein. Until recently, no reliable epidemiological data were available. The incidence is estimated to be higher than that of deep-vein thrombosis (DVT) (1/1000). SVT shares many risk factors...

Extended-Duration Treatment of Superficial Vein Thrombosis of the Lower Limbs with Tinzaparin

Purpose To identify risk factors for recurrent thromboembolic events (RTEs) and define the optimum duration of treatment with tinzaparin in patients with superficial vein thrombosis (SVT) of the lower limbs. Materials and Methods A total of 147 consecutive patients with significant SVT were treated with subcutaneously administered tinzaparin. The composite primary endpoint of the study was RT...

Regular Article CLINICAL TRIALS AND OBSERVATIONS Risk of venous and arterial thrombotic events in patients diagnosed with superficial vein thrombosis: a nationwide cohort study

Superficial vein thrombosis (SVT) is a relatively common condition of which the incidence was recently established to be about 0.6 per 1000 person-years (py). In the past, SVT has been considered a benign, self-limitingdisorder, requiring only symptomatic treatment. However, recent evidence showing that SVT is closely linked to occurrence of deep vein thrombosis (DVT) or pulmonary embolism (PE)...

BC Superficial vein thrombosis (SVT) is the preferred term to indicate thrombosis of any segment of the superficial vein system. SVT encompasses some older terms, such as superficial phlebitis (also called superficial thrombo phlebitis); suppurative thrombophlebitis (also called septic thrombophlebitis); and superficial femoral vein thrombosis (a misnomer). SVT also encompasses many current te...

A retrospective analysis of patients treated for superficial vein thrombosis.

INTRODUCTION The absolute risk of deep venous thrombosis (DVT) and pulmonary embolism (PE) as well as extension and/or recurrence in superficial vein thrombosis (SVT) of the leg is considerable and underestimated. We retrospectively evaluated therapeutic management, thrombophilic risk factors and clinical outcome of SVT. METHODS A database search was performed for consecutive patients with a ...
